In our laboratory, acoustic shearing with a Covaris instrument (Covaris, Woburn, MA) is typically done to obtain DNA fragments in the 100–5000 bp range, while Covaris g-TUBEs are employed for the 6–20 Kbp range necessary for mate-pair libraries. DNA fragmentation is typically done by physical methods (i.e., acoustic shearing and sonication) or enzymatic methods (i.e., non-specific endonuclease cocktails and transposase tagmentation reactions)( 12). Three approaches are available to fragment nucleic acid chains: physical, enzymatic, and chemical. The size of the target DNA fragments in the final library is a key parameter for NGS library construction. In general, the core steps in preparing RNA or DNA for NGS analysis are: ( i) fragmenting and/or sizing the target sequences to a desired length, ( ii) converting target to double-stranded DNA, ( iii) attaching oligonucleotide adapters to the ends of target fragments, and ( iv) quantitating the final library product for sequencing. Cell photograph courtesy of Annie Cavanagh, Wellcome Images. One difference between the two technologies is that Illumina allows sequencing from both ends of the library insert (i.e., paired end sequencing). The final step generates the actual sequence via the chemistries for each technology. The next step involves clonal amplification of the library, by either cluster generation for Illumina or microemulsion PCR for Ion Torrent. DNA Fragments are converted into the library by ligation to sequencing adapters containing specific sequences designed to interact with the NGS platform, either the surface of the flow-cell (Illumina) or beads (Ion Torrent). RNA is converted to cDNA by reverse transcription. RNA or DNA is extracted from sample tissue/cells and fragmented. Basic workflow for NGS library preparation. However, it should be noted that almost all of the principles discussed in this review can be applied with minimal modification to NGS platforms developed by Life Technologies, Roche, and Pacific Biosciences. ![]() Here, we compare and contrast various library preparation strategies and NGS applications, focusing primarily on those compatible with Illumina sequencing technology. ![]() For example, NGS library preparation has now been successfully demonstrated for sequencing RNA and DNA from single cells ( 3–11).įundamental to NGS library construction is the preparation of the nucleic acid target, RNA or DNA, into a form that is compatible with the sequencing system to be used ( Figure 1). During this time, as sequencing technologies have improved and evolved, so too have methods for preparing nucleic acids for sequencing and constructing NGS libraries ( 1, 2). ![]() Over the past five years, next-generation sequencing (NGS) technology has become widely available to life scientists. ![]() A 20-something girl from Michigan told me all about her job building transmissions. Among those attending were Adolf Hitler, Hermann Göring, Franz von Papen and Erich Koch, East Prussia's Nazi gauleiter.Ī year later, the monument again came to prominence on the death of Paul von Hindenburg. The Polish government allowed 1,500 cars to transit through the Polish Corridor. In August 1933 the Nazis held a massive demonstration at the memorial to commemorate the anniversary of the battle. The numbers of visitors did not meet expectations initially but during the Nazi era the numbers were such that the inn required to be extended. The architects had also built an inn nearby in traditional East Prussian style. Ten kilometers of veterans, resplendent in Imperial uniforms, paid homage to Hindenburg and the 20 unknown German soldiers from the 1914 battle who were interred at the memorial. An extract from the speech was later carved into a bronze plaque by the Nazi regime and installed in one of the towers of the memorial. His speech was deemed highly nationalistic and in keeping with the times for the Weimar Republic, but was not well received outside Germany since it denied German responsibility for the war. The 80-year-old Hindenburg was dressed in the uniform of a Colonel-in-chief of a Masurian regiment to which he'd been appointed by the Emperor (who had since abdicated). Opening and dedicationĪ gathering of thousands came to the dedication of the newly finished memorial on September 18, 1927. The design influenced other projects undertaken by architects and builders during the era. The monument's location on a hilltop was accentuated by massive earthworks and landscaping designed to look as if nature alone had shaped the site. The memorial was built in a prominent place in a shape reminiscent of the castles of the Teutonic Knights. The architects imagined the memorial to be a new volkish "community of the dead" and incorporated the burial of 20 unknown German soldiers from the Eastern Front into the project concept. This ideology was mooted in Germany in the 1920s and 1930s. In doing so, the architects anticipated the concept of Totenburgen (Fortresses of the Dead) housing mass graves of soldiers. The memorial embraced the Anglo/French concept of the Unknown Soldier. Concept & designĪerial view 1944, from a Luftwaffe plane. In the 1950s, Polish authorities razed the site, leaving few traces. As the Red Army approached in 1945, German troops removed Hindenburg's remains and partly demolished key structures. ![]() Adolf Hitler ordered the monument to be redesigned and renamed "Reichsehrenmal Tannenberg". When Reichspräsident Hindenburg died in 1934, his coffin and that of his wife, who had died in 1921, were placed there despite his wishes to be buried at his family plot in Hanover. The octagonal layout with eight towers, each 67 feet (20 m) high, was influenced by Holy Roman Emperor Frederick II's Castel del Monte, and by Stonehenge. The victorious German commander, Generalfeldmarschall Paul von Hindenburg, became a national hero, and was later elected Reichspräsident.ĭedicated by Hindenburg on the 10th anniversary of the Battle of Tannenberg in 1924 near Hohenstein (Ostpreußen) (now Olsztynek, Poland), the structure, which was financed by donations, was built by the architects Johannes and Walter Krüger of Berlin and completed in 1927. The Tannenberg Memorial was a monument to the German soldiers of the second Battle of Tannenberg in 1914 which was named after the medieval battle of the same name. In 1945, Hindenburg's coffin and several other items were removed from the site to protect them from advancing Soviet forces, and withdrawing German forces partially demolished the monument and collapsed several of the towers. The design in the center is an iron cross, a symbol originally from the Kingdom of Prussia but heavily used by Nazi Germany, and the whole site was designed to look like a medieval castle on a large hill.Īlthough it was initially intended to be a sort of tomb of the unknown soldier, in 1934 German general Paul von Hindenburg was buried there, and it became a site of pomp ceremony and military propaganda for Nazi Germany. This was the Tannenberg Memorial, built near the town of Hohenstein, East Prussia (now Olsztynek, Poland).īuilt in the mid-1920's, it was a monument dedicated to the German soldiers who fought in several battles in the area (two in WWI, and one from the 15th century). ![]() For more info, see Storyboarded animations. The Windows Runtime supports an interpolation logic so that you can animate from one Color to another in a From/ To animation and the animation will use interpolated Color values as the animation runs. These mostly support animating a Color value that exists on a Brush. There are also some XAML properties that take a direct Color value. You might also want to use system colors, which can be accessed as merged-in resources for themes that the system defines. For more info, see Optimize your XAML markup or ResourceDictionary and XAML resource references. If you use the same color brush often in your XAML, you should define a SolidColorBrush as a resource rather than using the inline implicit creation of new values, because that's more efficient. For more info on using brushes and colors in XAML, see Use brushes. This syntax implicitly creates a new SolidColorBrush with a Color value equal to Cyan that fills the Brush-type value of TextBlock.Foreground for that element. For example, you can set the Brush-type value TextBlock.Foreground using a syntax such as. ![]() However, the Brush type enables a XAML shorthand that lets you set an attribute value of type Brush using a named color string, or a format string that can be parsed into an ARGB form. Instead, a Color is used as a component value of a Brush (either SolidColorBrush or LinearGradientBrush). In most XAML UI scenarios, a Color isn't used directly as a property value of a UIElement. You can also use the Colors values for equality comparisons against a Color. For example, the Colors.AliceBlue property returns a Color that is equal to Color from the XAML usage. These properties are useful for setting Color values in code that match the attribute string form used in XAML to set a named Color. Various predefined Color values are available as static properties on the Colors class. ![]() ![]() A further issue with sending over glass slides is that the receiving department does not always send back all the slides because they need to archive the reviewed slides.Īnother logistical challenge involves pathology expert panels, where pathologists discuss difficult and/or interesting samples to reach consensus at a multiheaded microscope, requiring many pathologists to travel to a central laboratory with obvious loss of productivity. It also serves to translate the report into local usual and up-to-date language and ensures that material is available for demonstration at multidisciplinary meetings and for comparison with new material (eg, biopsies from metastases). Finally, during this process, patients are eagerly awaiting the final report with their diagnosis, treatments are delayed, and patient anxiety levels are high, often causing patients to call in sick.Ī similar process takes place when patients are referred to another hospital, in which case it is customary to ship the pathology material to reassess (revise) the patient’s situation because revision may take into account new information on the patient and hence the diagnosis may be adjusted or errors may be corrected. Patient reports can easily be taken from broken or opened packages during transportation, breaching patient privacy. In addition, it is also error prone slides may be broken during transportation or even get lost completely. This is an expensive and laborious logistic process that easily takes up to 2 weeks, depending on distances and adequacy of traditional mail logistics. Traditionally, such consultations take place by sending slides through mail back and forth. 1 Such specialists may not always be at hand locally, especially in smaller general practices, in which case a colleague in another practice can be consulted. Because erroneous diagnoses may lead to inadequate treatment of patients, it is often necessary to consult more specialized colleagues. In view of the explosion of knowledge in medical science, it has become impossible for the practicing pathologist to keep up with the developments in all fields and maintain sufficient diagnostic knowledge in every area. Pathology is a broad medical specialty covering cell and tissue diagnostics of all organs and body parts. Pathology Image Exchange was released in April 2018 after technical validation, and a first successful validation in real life has been performed for hematopathology cases. In this article, we describe the setup and validation of the Pathology Image Exchange project, which aimed to create a vendor-independent platform for exchange of whole-slide images between Dutch pathology laboratories to facilitate efficient teleconsultation, telerevision, and virtual slide panels. This poses high demands on the setup of such a platform, given the inherent complexity of the handling of digital pathology images. More importantly, a software platform is needed for exchange of these images and functionality to support the processes around discussing and reporting on these images without breaching patient privacy. This requires basic slide scanner infrastructure in participating laboratories to produce whole-slide images. Among the many uses of digital pathology, remote consultation, remote revision, and virtual slide panels may be the most important ones. ![]()
